Mechanism of Action

Orexins (hypocretins) are hypothalamic neuropeptides critical for maintaining wakefulness. Orexin-A (33 aa) activates both OX1R and OX2R receptors, while Orexin-B (28 aa) preferentially binds OX2R. They stabilise wake states, regulate feeding behaviour, and modulate reward pathways. Orexin neuron loss causes narcolepsy type 1.

Clinical Benefits

  • Regulates wakefulness and sleep-wake transitions
  • Modulates appetite and feeding behaviour
  • Supports energy homeostasis
  • Research applications in narcolepsy and sleep disorders
